5/6 nephrectomy-induced rat CKD model

The 5/6 nephrectomy model is a widely used experimental method to induce chronic kidney failure (CKD) in rats. By surgically removing 5/6 of the kidney mass, the remaining renal tissue undergoes hyperfiltration injury, leading to progressive glomerulosclerosis tubulointerstitial fibrosis, and renal dysfunction, mimicking human CKD progression.

Body weight curve of 5/6 nephrectomy-induced rat CKD model is shown in Fig. 2A. The 24h-urine volume, urine MALB and CREA level were significantly higher in 5/6 nephrectomy-induced CKD rats compared to the control group(Fig. 2B-D). The SBP, DBP and MBP were significantly higher in 5/6 nephrectomy-induced CKD rats compared to the control group at W6(Fig. 2E). Data were presented by mean ± SEM, n=4-8. *, p < 0.05; **, p < 0.01; ***, p < 0.001 and ****, p < 0.0001.

Histopathological analysis of the rat kidney demonstrated marked renal injury, characterized by diffuse inflammatory infiltrates, pronounced tubular dilation and tubular necrosis, extensive interstitial fibrosis, and irregular GBM thickening in 5/6 nephrectomy-induced CKD rats at W9.
