Introduction of NeoMab™ Mice

The NeoMab™ mouse retains native mouse constant region encoding genes while incorporating human variable gene repertoires into endogenous loci of BALB/c mouse. The generation of models employs site-specific knock-in techniques, wherein the human antibody variable region gene library is introduced into the endogenous antibody gene locus of the mouse while still retaining the mouse constant region gene segments. This strategy, also embraced by the NeoMab™ platform, overcomes the limitations associated with human gene segment numbers and preserves the regulation of endogenous expression and Fc-mediated signaling (Murphy, Macdonald, et al., 2014), resulting in an immune response closely resembling that of wild-type mice.

NeoMab™ mice utilize human V(D)J genes encoding antibodies, with a usage frequency and sequence diversity similar to that of humans. After immunization with human recombinant protein antigens, NeoMab™ mice display antigen-specific serum titers similar to BALB/c mice, reaching approximately 105-106after three immunizations.

Humanized Fab simplifies the development process and saves time. The antibody sequences obtained have undergone a natural antibody affinity maturation process within NeoMab™ mice, making affinity maturation more efficient.

The humanized antibody generated by NeoMab™ mice may exhibit lower immunogenicity. Antibodies obtained from NeoMab™ mice have high affinity (approximately 10-10-10-8 by SPR detection), excellent specificity, and minimal toxic side effects.
