Drug efficacy on high fat diet induced obesity model


Introduction:A diet-induced animal obesity model is established by feeding with high fat diet (HFD, Research Diets D12492) for 12-14 weeks. This obesity model can be used to test the effects of the drug on obesity and its related metabolic syndrome. 

Strain:C57BL/6J mouse and other gene editing mouse

Animal number:10-12mice per group,5-8 groups

Grouping:Negative control, Vehicle control, Positive drug control,Test drug (high, middle and low dose level).

Period: 16-20 weeks

Inspection ItemsBody weight,Food intake,Plasma glucose,HbA1c , GTT,ITT,Serum insulin level, Body fat ratio, Metabolic cage,Blood lipedema,Cytokine and chemokine assessment.

The body weight of C57BL/6J mice increased significantly after feeding with 60% high fat diet. Data were presented as Mean±SD. **, P<0.01; ***, P<0.001 compared to control using the unpaired two-tailed Student's t-test method.


2.Glucose Tolerance Test

Control and DIO mice were fasted for 6 hours and then gavage with 2g/kg glucose solution. Blood glucose was measured at 0, 15, 30, 60, 90 and 120 min after gavage. The glucose tolerance of DIO mice was significantly impaired. Data were presented as Mean±SEM. ***, P<0.001 compared to control using the unpaired two-tailed Student’s t-test method.


3. Serum insulin level

After 6 hours fasting, insulin levels were detected at 0 and 15 min in the oral glucose tolerance test. The results showed that insulin levels in DIO mice were significantly higher than those in the control group. Data were presented as Mean±SEM. ***, P<0.001 compared to control using the unpaired two-tailed Student’s t-test method.


4. Drug efficacy on the body weight of DIO mice

Orlistat significantly reduced the body weight of DIO mice. 10 mice per group. The data were presented as Mean±SEM.

Orlistat significantly reduced the body fat ratio of DIO mice after 28 days treatment. 10 mice per group. Data were presented as Mean±SEM. ***, P<0.001 using the one-way ANOVA with Dunnett post-hoc test method.
