Drug efficacy on type 2 diabetes (T2D) mouse model


Introduction:Leptin receptor deficiency mice (BKS-db mice) exhibit symptoms of T2D such as hyperglycemia and hyperinsulinemia from 4 weeks old. In addition, high fat diet feeding combined with low-dose Streptozotocin (STZ) injection could also induced the symptoms of T2D in C57BL/6J mice. These models are suitable for testing the anti-T2D drug efficacy. 

Strain:BKS-dbC57BL/6J mouse and other gene editing mouse

Group Size10-12mice per group,5-8 groups

Grouping:Negative control, Vehicle control, Positive drug control,Test drug (high, middle and low dose level).

Period: 8-12 weeks

Inspection ItemsBody weight,Food intake,Plasma glucose,HbA1c , GTT,ITT,Serum insulin level, Body fat ratio, Metabolic cage,Blood lipedema,Cytokine and chemokine assessment.

2.Drug efficacy on the plasma glucose of BKS-db

Test drug treatment was significantly reduced the plasma glucose of BKS-db mice. Control group n=14, BKS-db + PBS group n=12, BKS-db + Compound A group n=12. Data were presented as Mean ± SEM. **, P < 0.01; ***, P < 0.001 compared to the BKS-db + PBS group using the one-way ANOVA with Dunnett post-hoc test method.


3. Establishment of T2D by high fat diet feeding combined with low-dose Streptozotocin (STZ) injection

C57BL/6J mice were fed with 45% HFD and injected with low-dose STZ for 5 times. After 2 weeks of the first injection, the plasma glucose level of the model group was significantly higher than that of the control group. Data were presented as Mean ± SEM, n = 5 to 40. ****, P< 0.0001 using the unpaired two-tailed Student's t-test method.


4. Drug efficacy on the plasma glucose of HFD+STZ induced T2D model

The test drug significantly reduced the 6h fasting blood glucose and HbA1c levels of the T2D mouse model. Data are presented as Mean±SEM, n = 5 to 10. ****, P<0.0001 using the one-way ANOVA with Dunnett post-hoc test method.


5. Drug efficacy on the glucose tolerance of HFD+STZ induced T2D model

The test drug significantly improved the glucose tolerance of the T2D mouse model. Data are presented as Mean±SEM, n = 5 to 10. ****, P<0.0001 using the one-way ANOVA with Dunnett post-hoc test method.
